So consumables, in the mechanical sense, we're not ready to discuss as of yet. There is no planned change from lore usage/morality between versions of the game, however. That of course means that, whatever the mechanics of using poisons of any kind in game, they will still be considered the tools of evil people (such as Corathites or generally nasty scoundrels, for example).

Regarding lycanthropy, again, this isn't lore we're ready to release generally to the public. Much like vampire and Blood Seraph, lycanthropy has very specific lore on Layonara. This sort of information wouldn't be commonly known by even adventurers, a research project for them at needed times from guarded tomes and the like. At some point down the line we will likely be filling out some very basic information on the lore pages, like we did with the Blood Seraph, but probably as associated with quests.

In Mabinogi, part of the combat learning curve is knowing when to use certain combat skills. This can easily be broken down into a rock, paper, scissors match-up, in that certain skills are beaten by others. How much depth will the combat system in the Layonara MMO have?

Playing D&D 3.5e, D&D 5e, Pathfinder, and exploring Starfinder through a VTT

While I'm not really familiar (beyond the name) with Mabinogi, the new game's combat system isn't  presently intended to be anything of the sort that you describe (if I am understanding you properly), though as it is with many games, some skills and weapons may be more or less effective against a particular enemy.  There won't be any...say...use of Capo Ferro against Bonetti's Defense, countering that with Thibault, etc. There won't be any need to pick just the right move at the right time.

The depth of combat will come in with the Refined Combat Skills, which are in some ways like combat Feats in D&D/Pathfinder while being trainable like any other skill. In that regard, one's choice of which skills to attain and when/how they are used in combat could have a significant impact upon one's overall effectiveness in combat.
